Analytical Ultra Centrifugation combines an ultracentrifuge with optical absorbance measurements and monitors a sample sedimentation profile over time. This gives information on the samples overall shape, molar mass and size distribution.
What are the sample requirements?
- 1 mL of samples at concentrations between 0.5 and 1 mg/mL is sufficient for most analyses.
What other specific considerations are relevant?
- The technique relies heavily on selective absorbance measurements to monitor sedimentation effects so high levels of purity is not an absolute concern. However, this does become a factor of consideration if a specific absorbance wavelength is not selectable for the target protein in an impure mixture.
